Construct library reused across runs — kept in this browser Your constructs, kept between runs, with their default target OD, Agro strain, plasmid, tag, antibiotics and stock location.

Paste from a spreadsheet or a plain list — one construct per line, columns separated by tabs or commas. Then check the column each one lands in. A bare list of names works too. Rx:HA⇥GV3101⇥pICSL86977⇥HA⇥0.3

Construct Target OD Agro strain Plasmid Tag Antibiotics Stock Notes

Cultures culture mL = final vol × target OD ÷ (OD read × dilution) One row per culture. The OD you read and the dilution you read it at give the volume of neat culture, and the buffer that makes it up to the final volume.

Strain OD read Dilution Target OD Final mL On hand Culture Buffer Make-up Solo
OD read is the value off the spectrophotometer on the diluted sample — the dilution factor is applied for you. Solo counts a strain as its own infiltration in the prep summary; untick it for strains you only use inside mixes.

Mixes co-infiltration combinations One tube per co-infiltration. Each component is dosed against this tube's final volume and its own target OD; buffer tops the tube up.

One combination per line, components joined by +. Numbering like 1. is ignored. To name a mix yourself, put the name first and a tab or | before the components — otherwise it is named after what goes in it.

Construct summary each construct on its own Every construct as a single-strain tube: the neat culture and the buffer that brings it to its own final volume.

Combination summary what goes on the bench Every mix in one list: component count, buffer and final volume per tube, with the run totals.

Prep summary How much neat culture each construct needs across every mix it appears in, plus its own row if Solo is ticked, set against what you have on hand.

Each strain’s culture volume is final volume × target OD ÷ (OD read × dilution); buffer makes up the rest. In a mix, every component is dosed against the mix’s own final volume and its own target OD, and the buffer tops the tube up. Work is saved in this browser as you type.
